The effects of cadmium on zinc and copper in rat kidney and metallothionein in a dose-effect study

The questions as to the increase in zinc (Zn) and copper (Cu) contents of kidney and metallothionein in response to cadmium (Cd) injection were examined. Each rat was intraperitoneally injected once with 0.9% NaCl or cadmiun (1, 2 or 3 mg Cd/kg b.w.). After Cd injection, 61.8 – 64.0% of the renal Cd contents, 62.5 – 65.1% of the renal Zn contents and 61.3 – 65.2% of the renal Cu contents were found in the cytosol. The Cd contents in the cytosol and kidneys increased in response to Cd injection. Although the Zn and Cu contents in the cytosol and kidneys of Cd-injected rats were higher than those of control rats, they did not increase in response to Cd injection. The distribution profiles of the renal cytosol of Cd-injected rats on a Sephadex G-75 column showed that the amount of the increased Cd was attributable to the metallothionein (MT) fractions. The Cd concentrations in the MT fractions increased in response to Cd injection. A dose-dependent increase in the Zn and Cu concentrations of the MT fractions was not observed in kidneys with increasing doses of Cd. These results indicated that Cd injection could influence Zn and Cu concentrations in rat kidneys and in the MT fractions.Correspondence to:

Efficacy of H2 receptor antagonists for prevention of upper gastrointestinal bleeding during dual-antiplatelet therapy

Proton pump inhibitors (PPI) are frequently used to prevent upper gastrointestinal bleeding (UGIB) in patients receiving dual-antiplatelet therapy (DAPT) with aspirin and clopidogrel. However, the concomitant therapy of PPI and DAPT has been associated with a decreased effect of the antiplatelet drugs and an increased risk of major adverse cardiovascular events (MACE). It has been suggested that histamine H2 receptor antagonists (H2RA) can be used as alternatives to PPI to prevent UGIB during DAPT without an increase in the risk of MACE. We tested this hypothesis in a retrospective cohort study including patients without a prior history of upper gastrointestinal events. We examined the incidence of UGIB and MACE in 296 patients treated with H2RA (H2RA group) and 447 patients not treated with H2RA (control group) during DAPT with aspirin and clopidogrel after drug-eluting stent implantation. The patients treated with PPI were excluded. In the 1-year follow-up, UGIB occurred in 2 patients (0.7%) in the H2RA group and 12 (2.7%) in the control group. The incidence of UGIB was significantly different between the two groups (p = 0.049 in log-rank test). MACE occurred in 31 patients (10.5%) in the H2RA group and in 54 patients (12.1%) in the control group, and the incidence was not significantly different (p = 0.447 in logrank test). Thus, H2RA may be effective safe alternatives to PPI during DAPT in patients without a prior history of upper gastrointestinal events.Correspondence to:

The effect of zinc on copper in rat kidney, liver, and in metallothionein in a dose-effect study

In the present study I examined the effect of zinc (Zn) on copper (Cu) contents of kidneys, livers and metallothionein (MT) after Zn injection. Each rat was intra-peritoneally injected once with 0.9% NaCl or Zn (5, 10, or 20 mg Zn/kg BW). After Zn injection, 65.2 – 74.8% of the hepatic Zn contents, 61.0 – 63.0% of the renal Zn contents, 64.9 – 68.1% of the hepatic Cu contents, and 62.3 – 65.7% of the renal Cu contents were found in the cytosol. The Zn contents in the hepatic cytosol, the renal cytosol, livers, and kidneys increased in response to Zn injection, while no difference in the Cu contents of the hepatic cytosol and livers, was observed between the control rats and Zn-injected rats. The Cu contents of the renal cytosol and kidneys showed a decrease with increasing doses of Zn. The distribution profiles of the renal cytosol of Zn-injected rats on a Sephadex G-75 column showed that this decrease was correlated with Cu concentrations of the MT fractions. The Cu contents in the renal MT fractions of rats injected with 20 mg Zn/kg BW decreased to ~ 80% of the control rats. These results indicated that Zn injection could influence Cu contents in rat kidneys, in the renal cytosol, and in the MT fractions, but it had no effect on Cu contents in rat liver and in the hepatic cytosol.Correspondence to:

The effect of cadmium on zinc in rat liver and metallothionein in a time-course study

In the present study I examined the effect of cadmium (Cd) on zinc (Zn) contents of livers and metallothionein (MT) after Cd injection. Each rat was intraperitoneally injected once with 2 mg/kg b.w. of Cd as CdCl2. The rats were killed after 7, 14, and 21 hours. The contro rats were untreated. After Cd injection, 70.6 – 82.3% of the hepatic Cd contents and 60.9 – 63.2% of the hepatic Zn contents were found in the cytosol. The Cd and Zn contents in the hepatic cytosol and the livers increased during the time (0 – 21 hours). The distribution profiles of the hepatic cytosol of Cd-injected rats on a Sephadex G-75 column showed that the increased Cd and Zn were attributable to Cd and Zn concentrations of the MT fractions, respectively. The Cd and Zn concentrations in MT fractions within 21 hours increased to a maximum 21h. These results indicated that Cd injection could influence Zn contents in rat livers, in the cytosol and in the MT fractions in a timecourse study.Correspondence to:

The effect of silver on zinc and copper in rat liver and metallothionein in a time-course study

Each rat was intraperitoneally administered once with 20 mg/kg b.w. of Ag as AgNO3. The rats were killed at 7, 14, and 21 hours. The control rats were untreated. After Ag injection, 63.3 – 70.5% of the hepatic Ag contents, 53.1 – 60.1% of the hepatic Zn contents and 54.9 – 63.3% of the hepatic Cu and were found in the cytosol. The Zn and Cu contents in the hepatic cytosol and the livers increased during the time (0 – 14 hours), and remained unchanged thereafter. The distribution profiles of the hepatic cytosol of Ag-injected rats on a Sephadex G-75 column showed that the increased Ag, Zn, and Cu were attributable to Ag, Zn, and Cu concentrations of the MT fractions, respectively. The Ag, Zn, and Cu concentrations in MT fractions increased during the time (0 – 14 hours) and remained unchanged thereafter. The effect of copper on zinc in rat kidney and metallothionein in a dose-effect study

Each rat was injected intraperitoneally once with 0.9% NaCl or Cu (2, 4, or 6 mg Cu/kg body weight (BW)). After Cu injection, 60.0 – 63.3% of the renal Cu contents and 62.4 – 66.2% of the Zn contents were found in the cytosol. The Cu contents in the cytosol and kidneys increased following the injected amounts in the range of Cu dose of 0 and 4 mg kg BW, and remained unchanged thereafter. Although Zn contents in the cytosol and kidneys of the Cu-injected rats were higher than those of the control rats, they did not increase in response to Cu injection. The distribution profiles of the renal cytosol of Cu-injected rats on a Sephadex G-75 column showed that the amounts of the increased Cu and Zn were attributable to the MT fractions. The Cu concentrations in the MT fractions increased in response to Cu injection. It is interesting that in spite of no change in the Zn contents of the MT fractions with increasing doses of Cu, the Zn concentrations in the MT fractions of Cu-injected rats were higher than the control values. These results indicated that Cu injection could influence Zn contents in rat kidneys, the cytosol and the MT fractions in a dose-effect study.Correspondence to:

Mechanisms of metabolic disorders of Cu and Zn in rat kidney after Au injection: A review

Gold (Au) is a heavy metal, and its complexes are one of the more effective medicaments available for rheumatoid arthritis, although many authors have reported serious kidney damage during chrysotherapy. In this article, the mechanisms of the DNA damage and metabolic disorder of copper (Cu) and zinc (Zn) in renal kidney of rats after Au injection are reviewed. Cu and Zn contents in the kidney of the Au-injected rats dramatically increased in comparison with the control rats, and Cu-binding metallothionein (MT) was predominantly observed in the outer stripe of the outer medulla in the kidney. The signals of terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-biotin nick end labeling (TUNEL) and the immunoreactivity of 8-hydroxydeoxyguanosine (8-OHdG) were located in the cortex of the Au-injected rats. On the other hand, there were yellow and orange fluorescent signals in the renal cortex of rats administered with Cu. These findings suggested that the oxidative DNA damage in the kidneys of rats injected with Au is associated with Cu of the cortex except Cu-binding MT (Cu-MT) and the differential localization of renal Cu-MT induced by Au and Cu injection to rats. I postulated that the DNA cleavage might be caused by hydroxyl radicals formed by monovalence of Cu, that the mechanism of Cu accumulation in the kidney after Au injection could be different from that of Cu accumulation in the kidney after Cu injection, and that the high Cu accumulation in the kidneys of rats injected with Au was considered to be caused by dysfunctional ATP7B (a type of genes encoding P-type cation-transporting ATPase), because MT localization in the kidneys showed the same pattern in the kidneys of Long-Evans Cinnamon (LEC) rats as an animal model for Wilson’s disease. I also postulated that the high Zn accumulation in the kidneys of rats injected with Au was considered to be caused by dysfunctional ZnT (Zn transporter).Correspondence to:
