Vascular disrupting agents in non- small cell lung cancer

Abstrakt

The importance of a functional vascular supply for tumor growth and metastasis has been generally accepted. Vascular disrupting agents (VDAs) have been designed to particularly target preexisting blood vessels which may lead to a vascular shut-down. As a consequence, tumor vessels are no longer able to supply surrounding tumor cells with oxygen and nutrients which results in extensive tumor necrosis in preclinical studies. Results of clinical phase I and II studies indicate that VDAs may be active drugs in non-small cell lung cancer (NSCLC). In the following review the mechanisms of action of VDAs are described in comparison to anti-angiogenic therapy as well as possible combinations of VDAs with cytotoxic or anti-angiogenic agents.
